2018, Cytokine and Growth Factor ReviewsCitation Excerpt :Whereas GATA2 expression is tightly regulated during hematopoiesis, overexpression of GATA2 is frequently observed in AML patients where it is linked to poor prognosis [40]. Thus, enhanced IL-1-induced p38-signaling, as observed in IL-1β-sensitive AML patients, may result in the dysregulation of GATA2 activation in the context of leukemic cell proliferation and thereby be a crucial factor in AML disease progression [17,39,41–43] (Fig. 1). Because normal hematopoietic stem cells (HSCs) experience severe impairment of self-renewal capacity and even undergo growth suppression upon IL-1β exposure, whereas IL-1β acts as a potent survival factor for leukemic cells [11], IL-1β and components of the IL-1β signaling pathway may be promising targets for future therapeutic approaches.
Interleukin-1β as emerging therapeutic target in hematological malignancies and potentially in their complications
2017, Blood ReviewsCitation Excerpt :This review presents the pathogenic role of dysregulated IL-1β in patients of hematological malignancies, its promising therapeutic value in preclinical models, and its potential contribution linking to second disease and complications based on lessons learned from other systemic inflammatory diseases. IL-1β is mainly produced by myeloid cells [10,11]. It is synthesized as an inactive form (Fig. 1A), pro-IL-1β that is activated intracellularly by caspase 1 [8,11].
Psychological and cytokine changes in children and adolescents undergoing hematopoietic stem cell transplantation
2007, European NeuropsychopharmacologyCitation Excerpt :Various confounding factors may affect cytokine production and activity. These factors include stress (Raz, 1997), steroid treatment (Bagby, 1989; Oppenhiem and Ruscetti, 2001), Granulocyte Colony-Stimulating Factor (G-CSF) administration (Oppenhiem and Ruscetti, 2001; Tayebi et al., 2001; Vilcek, 1998), and the use of antibiotics. The diagnoses of our subjects are heterogeneous leading to different protocols and intervals of time in the transplantation process.
Monocyte-derived CXCL7 peptides in the marrow microenvironment
2006, BloodCitation Excerpt :In vitro, the monocytes, which arise from the hematopoietic stem cell, can form up to 40% of total cells in LTCs despite serial passages.7 Monocytes/macrophages are known to secrete large quantities of several cytokines, including IL-1, IL-6, G-CSF, TNF, PDGF, TGF-β, and M-CSF, all of which can affect both stromal cells and hematopoietic cells.8-13 Increasing proportions of macrophages in primary LTCs are associated with decreasing numbers of hematopoietic progenitors.14,15
